The Research Question . In 2017, a team of researchers in France reported that they had found a potential biomarker of the reservoir in the blood—a protein called CD32. In their attempts to replicate the discovery, several other research teams found that CD32 may be a marker of HIV activity in blood cells rather than a specific marker of the latent reservoir. amfAR INSTITUTE FOR HIV CURE RESEARCH. Established in 2016 with a five-year $20 million grant to the University of California, San Francisco, the Institute is the cornerstone of the Countdown to a Cure. Its teams of scientists work across the research continuum—from basic science to clinical studies—in a collaborative venture involving UNCOVERING NOVEL ANTI-HIV FACTORS Research Question Elite controllers, capable of suppressing HIV replication in the absence of antiretroviral therapy (ART), represent less than 1% of all HIV-positive individuals. While small in number, their participation in research studies has been critical to our understanding of how a DEPLOYING NEW IMAGING TECHNIQUES FOR HIV Findings and Impact. Dr. Timothy Henrich and colleagues from the University of California, San Francisco, reviewed a “toolbox” of nuclear imaging approaches—technology that has demonstrated success in assessing the spread of cancers—to directly visualize HIV and its impact on inflammation and metabolism throughout the body. MOVING CLOSER TO A CURE: REDUCE AND CONTROL STRATEGIES The Research Question. CAR T cells against HIV were among the earliest CAR T cells developed. However, the greatest strides in the clinic have been made by deploying these cells to recognize and target certain cancers—the FDA approved two CAR T cell therapies againstcancer in 2017.
MAPPING AN HIV VACCINE TARGET Researchers studied the region of HIV recognized by a bNAb obtained from an elite neutralizer. They used highly sophisticated scanning procedures such as ultra-high resolution cryo-electron microscopy—which examines the topography of proteins—to capture the molecular interactions between the antibody and the participant’s own HIV. IN THE VANGUARD OF VACCINE RESEARCH amfAR grantee Dr. Dan Barouch is director of the Center for Virology and Vaccine Research at Boston’s Beth Israel Deaconess Medical Center. In observance of HIV Vaccine Awareness Day on May 18, amfAR Vice President and Director of Research Dr. Rowena Johnston spoke to Barouch about the key challenges in vaccine development, the pursuit of vaccines to potentially cure HIV, and why we should COULD GENE THERAPY CURE HIV? CTLS: STORMTROOPERS OF THE IMMUNE SYSTEM CTLs, also known as CD8+ T cells, are cells of the immune system that specialize in recognizing and killing virus-infected cells.
